AstraZeneca Highlights P-III (SERENA-6) Trial Data on Camizestrant Combination for ESR1 Mutated Advanced Breast Cancer at ASCO’26

- AZ‘s trial assessed camizestrant (n=157) vs AI (anastrozole/letrozole; n=158), both in combination with CDK4/6 inhibitor in 315 pts with LA/M HR+/HER2- advanced breast cancer harboring ESR1 mutation; filings supported by the trial are under review in the US, EU & Japan
- Trial showed 24mos. PFS rate of 34.9% vs 14.2% (mPFS: 16.8 vs 9.2mos.) & 24mos. time to second disease progression (PFS2) rate of 50.8% vs 36.3% (mPFS2: 25.7 vs 19.1mos.), with OS rate of 29.3% vs 31% (OS maturity: 30%; under evaluation)
- Pts switched to camizestrant showed a 99% median reduction in total ctDNA by Wk. 8, with 51% achieving total ctDNA clearance, vs a 64% increase & 1.9% clearance. Total ctDNA clearance was linked with improved OS; therapy also delayed the need for CT or ADCs (22.6 vs 18.7mos.) & improved PROs
